Photometric Detection of Nicotinamide Adenine Dinucleotides

چکیده

was first discovered by Arthur Harden and William Youndin in 1906. It plays key roles as a carrier of electrons and a participant in metabolic redox reactions, as well as in cell signaling. There are two forms of this coenzyme in cells, the non-phosphorylated form NAD + and the phosphorylated form NADP + . These two related coenzymes have similar chemistry, but perform different roles in metabolism. When both coenzymes are being discussed, they are referred to collectively as NAD(P) + .

Bioluminescent cell-based NAD(P)/NAD(P)H assays for rapid dinucleotide measurement and inhibitor screening.

Abstract The central role of nicotinamide adenine dinucleotides in cellular energy metabolism and signaling makes them important nodes that link the metabolic state of cells with energy homeostasis and gene regulation. In this study, we describe the implementation of cell-based bioluminescence assays for rapid and sensitive measurement of those important redox cofactors. We show that the sensit...

Effects of the Calvin cycle on nicotinamide adenine dinucleotide concentrations and redox balances of Xanthobacter flavus.

The levels of reduced and oxidized nicotinamide adenine dinucleotides were determined in Xanthobacter flavus during a transition from heterotrophic to autotrophic growth. Excess reducing equivalents are rapidly dissipated following induction of the Calvin cycle, indicating that the Calvin cycle serves as a sink for excess reducing equivalents. The physiological data support the conclusion previ...

Preparation and properties of some nicotinamide adenine dinucleotide analogues with pentose and purine modifications.

The structure of nicotinamide adenine dinucleotide was proved conclusively by chemical synthesis (1) and isolation (2) of the biologically active coenzyme from a reaction in which the 5,5’dinucleoside pyrophosphate was formed by condensing together the two component 5’-mononucleotides, adenosine monophosphate and nicotinamide mononucleotide, with dicyclohexylcarbodiimide or trifluoroacetic acid...

Ruling out pyridine dinucleotides as true TRPM2 channel activators reveals novel direct agonist ADP-ribose-2′-phosphate

Transient receptor potential melastatin 2 (TRPM2), a Ca(2+)-permeable cation channel implicated in postischemic neuronal cell death, leukocyte activation, and insulin secretion, is activated by intracellular ADP ribose (ADPR). In addition, the pyridine dinucleotides nicotinamide-adenine-dinucleotide (NAD), nicotinic acid-adenine-dinucleotide (NAAD), and NAAD-2'-phosphate (NAADP) have been shown...

Oxidation of reduced nicotinamide adenine dinucleotide phosphate by isolated corn mitochondria.

Isolated corn (Zea mays L.) mitochondria were found to oxidize reduced nicotinamide adenine dinucleotide phosphate in a KCl reaction medium. This oxidation was dependent on the presence of calcium or phosphate or both. Strontium and manganese substituted for calcium, but magnesium or barium did not. The oxidation of NADPH produced contraction of mitochondria swollen in KCl. Further evidence tha...
